Adult Stem Cells for Tissue Engineering

 

Tissue engineering-based treatments are beginning to be employed in the clinic for the treatment of an array of important clinical problems. For this purpose, autologous mesenchymal stem cells (MSCs) that can be readily isolated bone marrow, fat or other tissues offer advantages over differentiated cells that need to be isolated from tissue harvested from the patient, with attendant donor site morbidity. Our ongoing studies are employing an array of methods to identify MSC populations: evaluation of the percentage of cells expressing selected surface markers by flow cytometry; gene profiling using DNA microarray analysis; and bioassays. One goal is to correlate specific characteristics of the MSCs assessed in vitro with their capability to regenerate tissue in vivo.
